Chromatography trace for protein purification RD4 SERIES MONITORS

UV MONITOR FOR PREPARATIVE CHROMATOGRAPHY and FPLC

Simultaneous observation of UV Absorption at 280 and 250nm, Refractive Index and Conductivity

The first chromatography monitor with UV LED light sources and 0.001-800 AU absorption range

UV LED light sources provide over a decade of consistent function while eliminating warm-up time. UV light intensity is temperature-compensated to minimize long-term drift.

Demonstration of Absorption Range 0.001-800 AU at 10mm

Five 1ml samples of mouse mAb in PBS were sequentially eluted through the detector.

The samples contained 0.0001 to 10 mg of the antibody.

EXCELLENT MONITOR FOR PROTEIN PURIFICATION

THE ONLY CHROMATOGRAPHY DETECTOR FOR ORGANIC AND TOTAL SYNTHESIS

CONVENIENT ALTERNATIVE TO FRACTION COLLECTORS

Up to 16 solenoid valves can be added to all detectors. This allows to automatically collect fractions of unlimited volume into user-supplied vessels. The collection criteria are based on signal thresholds (UV absorption, RI, conductivity or pH), time windows, or a combination of both.

TECHNICAL CHARACTERISTICS

LIQUID CONNECTIONS

1/4"-28TPI compression fitting to 1/16" or 1/8" OD plastic tubing

OPERATION MODE

  • Autonomous: all unit functions are controlled from the front panel, no external PC is needed.
  • Five parameters are continuously recorded to the detector non-volatile memory.
  • Up to four out of five parameters can be selected at any time by user to be plotted as real-time graphs on the color LCD.

LIGHT SOURCES

  • 280nm and/or 250nm UV LEDs, followed by narrow bandpass diffraction filters
  • Spectral bandwidth: 10nm
  • Wavelength accuracy: ± 3nm
  • Current through UV LEDs: ≈ 10% of the maximum allowed for the corresponding UV LED. The operating current reading is available on the detector's screen.
  • Thermally compensated to minimize long-term drift
  • Typical lifetime: 15 years of non-stop operation

FLOW CELL

  • A single REACH Devices wide-range embedded flow cell is implemented, US Patent 8570521.
  • Total flow cell volume: 50μl
  • Flow rates: 0.1 to 300ml/min, no split - all eluants travel through the flow cell via the same path.
  • Maximum operating pressure: 1000 psi (68 bar)
  • Wetted materials: quartz, sapphire, PEEK, titanium, FFKM (Kalrez)
  • Solvents compatibility: all except HF, concentrated H2SO4, concentrated HNO3, and molecular halogens
  • Long term pH stability: 0-13
  • Hydrodynamic resistance: low, see graph

UV MONITORING

  • Range: 0.001 – 800 AU (Absorption Units)
  • Resolution: 0.0001 AU
  • Baseline noise: < 0.0002 AU
  • Drift: 0.0003 AU/hr at constant temperature (dry flow cell)

CONDUCTIVITY MONITORING

  • Two bright platinum electrodes
  • 0.01-150 mS/cm range

REFRACTIVE INDEX MONITORING

  • Measurement type: absolute
  • Measuring wavelength: 280nm
  • Range: 1.2000 to 1.7000
  • Resolution: 0.0001
  • Thermally compensated measurements

POWER OUTAGE

The recorded data is not corrupted. The detector will automatically resume data recording after the power is back on.

ACQUISITION RATES (non-stop run times)

  • 5Hz (2 hour 38 min)
  • 2Hz (6 hours 37 min)
  • 1Hz (13 hours 14 min)
  • 0.5Hz (26 hours 28 min)
  • 0.2Hz (66 hours 10 min)
  • 0.1Hz (132 hours 20 min)

DATA TRANSFER

By user's command to USB drive. No data acquisition occurs during data transfer.

COMMUNICATION INTERFACE

  • 4 independent analog outputs in 0-1V range (see manual for transfer functions), corresponding to (a) UV 250 nm, (b) UV 280 nm, (c) refractive index, (d) conductivity channels. The output (a) can be reassigned by a user to provide data from any channel and be used to provide the respective analog signal for contemporary fraction collectors.
  • One digital input pin for fraction count pulse (e.g. from a fraction collector). Detector can react to intermittent mechanical contact closure and TTL level signal.
  • Two digital outputs which can be programmed by REACH Devices as requested by the user
  • All input and output channels are galvanically isolated from mains ground and detector's internal circuitry ground for safety and noise resistance.
  • Up to two relays which can be set by the user to react on change in UV absorption, RI or conductivity.
